MOA (mechanism of action):

Amoxicillin hinders the synthesis of peptidoglycan – a crucial component of the cell wall of Gram-positive and a minor component of Gram-negative bacteria, thus, causes the bacteria death. Clavulanic Acid in its turn deactivates a beta-lactamase enzyme produced by some bacteria to hinder the action of Amoxicillin.

Amoxicillin is effective against gram-positive bacteria including Staphylococcus spp. (except strains producing penicillinase), Streptococcus spp.; aerobic gram-negative bacteria including Neisseria Gonorrhoeae, Neisseria Meningitidis, Escherichia Coli, Shigella spp., Salmonella spp., Klebsiella spp.

Recommended adult dose is 1200 mg thrice (every 8 hours).

In more serious infections frequency intervals should be increased to every 6 hours.

Treatment should not continue more than 14 days without review.

The duration of therapy should be determined by the response of the patient.

Certain infections like osteomyelitis may require a longer treatment.

For surgical prophylaxis in adults and children over 40 kg:

For procedures under 1-hour duration: 1200 mg given at the induction of anesthesia.

For procedures over 1-hour duration: 1200 mg given at the induction of anesthesia, with up to three doses of 1200 mg within 24 hours.

For procedures with a high risk of infection (e.g., colorectal surgery): 1200 mg three/four times within 24 hours.

These doses are usually given every 8 hours.

This regimen can be continued for several days if the procedure has a significantly increased risk of infection.
